Introduction:
Rabeprazole, also known as Pariet, is a proton pump inhibitor medication that is used to reduce stomach acid production in the treatment of various gastrointestinal disorders. Rabeprazole works by inhibiting the activity of proton pumps in the stomach lining, which in turn reduces the amount of acid that is produced. This medication is available in various forms, including tablets and capsules, and is prescribed by doctors for a range of conditions such as GERD, stomach ulcers, and Zollinger-Ellison syndrome.

How to Consume:
Rabeprazole should be taken as per prescribed by the doctor.
The dosage form and quantity of drug might be different for every acid related disorder:
For Peptic Ulcer:
The standard oral recommended dose for peptic ulcer is 20mg once daily after morning breakfast for upto 4 weeks.
Stomach infections ( caused due to Helicobacter pylori-bacteria):
The standard oral recommended dose for Stomach infections is 20mg twice a day after morning and evening meals for upto 7 days
For Helicobacter pylori-bacteria infection :
Rabeprazole is taken in combination with 1000mg(1g) Amoxicillin and 500mg Clarithromycin similarly twice a day for 7 days course.
Gastroesophageal Reflux Disease (GRED) –
The standard oral recommended dose for GRED is 20mg once daily for 4-8 weeks; if the patient is still not healed after 8 weeks, a further 8-week treatment may be considered.
Hyper secretory conditions (Zollinger-Ellison Syndrome)
The standard oral recommended dose for Hyper secretory conditions is 60mg once daily according to the severity it can also be 100mg a day or 60mg every 12hrs.
Storage Conditions:
Keep the medication in a tightly closed container, and out of reach of children. Store it at room temperature and away from excess heat(sunlight) and moisture.
Duration Between Two Dose:
The duration between each dose of medication varies depending on the specific disease being treated. For acid-related disorders, the standard dose is typically administered once or twice per day. However, for certain conditions, such as those requiring more intensive treatment, the duration between doses may be extended to 12 hours.

Drug Mechanism:
Pharmacodynamics:
Rabeprazole works by inhibiting the proton pump in the parietal cells of the stomach. The proton pump is responsible for producing acid in the stomach. By inhibiting the proton pump, rabeprazole reduces the amount of acid produced in the stomach, leading to acid suppression.
Rabeprazole reduces the amount of acid produced in the stomach by up to 95%. It works more rapidly than other PPIs, with effects seen within hours of ingestion
Rabeprazole binds covalently to the proton pump molecule, preventing it from producing acid. This binding is irreversible and lasts for up to 24 hours, allowing for long-term acid suppression.
Pharmacokinetics:
Rabeprazole is typically administered orally in the form of a tablet. It is rapidly absorbed by the body, with peak plasma concentrations occurring within 3-4 hours of administration.
After absorption, rabeprazole is primarily bound to plasma proteins in the blood. Approximately 97-98% of the drug is bound to proteins, which can affect its distribution to tissues and organs.
Rabeprazole has been shown to have a low volume of distribution, meaning that it is primarily distributed to tissues with a high blood supply such as the liver, kidney,
and gastrointestinal tract. It has also been found to cross the blood-brain barrier, although the significance of this is not well understood.
Rabeprazole undergoes extensive metabolism in the liver, primarily by the cytochrome P450 (CYP) enzyme system.
Rabeprazole and its metabolites are excreted mainly in the urine, with a small amount being eliminated in the feces.

Child Dosage:
· Gastroesophageal Reflux Disease (GRED) –
Below 12 Years:-
The dose for children below 12yrs as per prescribed by the doctor.
12years and Above
The standard oral recommended dose for children’s 12yrs and above is 20mg once daily for up to 8weeks

Important Note:
